Single excitation band fiber photometry measurements use an FMC with 3 ports: one port for the excitation light, one for the fluorescence detection, and one to connect to the sample. The cube has a dichroic mirror to let the excitation light through and separate the fluorescence emission and narrow bandpass filters to limit the excitation or fluorescence spectrum. E stands for fluorescence excitation, F for fluorescence, and S for the sample port on the cube's schematic. Currently, only Green Fluorophore (GFP-like) configurations or Red Fluorophore (RFP-like) are offered. Still, the filter set can be customized for other fluorophores on request.

The Intensity Adjustment Ring on the 460-490 nm LED light source reduces the source power below the LED driver's range if needed for low-power fiber photometry experiments.

Compared to Fluorescence Mini Cubes with only FC connectors on all ports, the ones with integrated photodetectors provide a 100% better signal-to-noise ratio (30% signal increase and a 40% noise reduction). Moreover, the versions where LEDs are integrated as well reduce the number of optical connections and the system’s overall footprint.
